سابقه و هدف: اسید اسکوربیک (Ascorbic acid) به وفور در مغز وجود داشته و بر تولید و انتقال میانجی های عصبی اثر می گذارد. در انتقال درد، سیستم گلوتاماترژیک نقش دارد. حضور اسید اسکوربیک گیرنده های گلوتاماتی NMDA را مهار و به دلیل اسیدیتهی آن، TPRV1 و ASIC را فعال می کند. لذا بررسی اثر تجویز درون صفاقی (i.p) و یا نخاعی (i.t) اسید اسکوربیک و اسکوربات سدیم بر احساس درد، هدف این مطالعه است.
مواد و روش ها: موش های صحرایی نر بالغ ویستار (250-200 گرم) در هشت گروه هفت تایی دریافت کننده ی سالین (i.p)، سالین (i.t)، اسید اسکوربیک ( mg/kg، i.p1،10،100،200)، اسید اسکوربیک (µl، i.t10/mg 02/0) معمولی و خنثی (اسکوربات سدیم) استفاده شدند. آستانه ی درد، توسط آزمون Tail-flick (TF) و شدت احساس درد شیمیایی طی آزمون فرمالین تعیین شد.
یافته ها: اسید اسکوربیک (mg/kg، i.p 10،100،200) در آزمون TF، پردردی (01/0P<) و در آزمون فرمالین، کاهش درد (05/0P<) ایجاد کرد. اسید اسکوربیک داخل صفاقی با دوز mg/kg 1 در آزمون TF فاقد اثر بود اما درد مرحله بینابینی آزمون فرمالین را کاهش داد (01/0P<). در تجویز نخاعی، اسید اسکوربیک با غلظت µl10/mg02/0 در آزمون TF موجب پردردی شد (001/0P<)، ولی به صورت اسکوربات سدیم، بی اثر بود. تجویز نخاعی اسید اسکوربیک و اسکوربات سدیم، درد را در آزمون فرمالین کاهش دادند (001/0P<).
نتیجه گیری: پردردی ناشی از تجویز درون صفاقی اسید اسکوربیک در آزمون TF، شاید حداقل بخشی به دلیل اثر مهاری کمتر آن بر گیرنده های AMPA و در تجویز نخاعی علاوه بر آن، حساسیت گیرنده های TRPV1 و کانالهای ASIC به اسیدیته ی اسید اسکوربیک باشد، زیرا تجویز اسکوربات سدیم اثر پردردی از خود نشان نداد. احتمالا اثرات مهاری اسید اسکوربیک و یا اسکوربات -سدیم طی تجویز درون صفاقی و نخاعی، بر گیرنده های NMDA و کاهش رهایش Substance P و دیگر عوامل التهابی در بروز بی-دردی طی آزمون فرمالین نقش داشته اند.
Background and Aim: Ascorbic acid (ASC) is abundant in the brain. ASC is effective on synthesis and transportation of neurotransmitters. Glutamatergic system is involved in pain transduction. NMDA receptors can be inhibited، and TPRV1 and ASIC receptors are excited due to its acidity at the presence of ASC. Therefore، this study is aimed to investigate the effects of intraperitoneal (i.p) and intrathecal (i.t) administration of ASC and sodium-ascorbate on pain.
Materials and Methods: Male Wistar rats (200-250 g) in eight groups (n=7) included saline (i.p)، saline (i.t)، ASC (1، 10، 100، 200 mg/kg، i.p)، usual and neutralized ASC (0.02 mg/10 μl، i.t) were used. Pain threshold was measured by tail-flick test and chemical pain was assessed using formalin-test.
Results: ASC (10، 100، 200 mg/kg، i.p) induced hyperalgesia in tail-flick test (P<0.01) and reduced pain in formalin-test (P<0.05)، but ASC (1 mg/kg i.p) with no effect in tail-flick، decreased pain during formalin-test interphase (P<0.01). Intrathecal ASC at 0.02 mg/10 µl concentration، but not sodium-ascorbate، induced hyperalgesia in tail-flick test (P<0.001). ASC (0.02 mg/10 µl، i.t) in both forms of usual and sodium-ascorbate، decreased pain in formalin-test (P<0.001).
Conclusion: Lesser inhibition effects of ASC on AMPA receptors may be responsible at least in part for its hyperalgesia in tail-flick test at i.p administration. This plus the sensitivity of TRPV1 and ASIC receptors to ASC acidity can be considered for i.t administration، because sodium-ascorbate (i.t) didn’t induce hyperalgesia. Inhibiting effects of i.p and i.t administration of ASC and sodium-ascorbate on NMDA receptors، release of SP and other inflammatory mediators، were probably some players of pain reduction during the formalin-test.
