خلاصة:
سابقه و هدف: با وجود ارزش بالای گوشت طیور، کنترل و بازرسی دقیقی بر روی لاشه های ذبح شده طیور در کشتارگاهها صورت نمی گیرد. در نتیجه امکان انتقال برخی از باکتری ها مانند اشریشیاکلی که یکی از عمده ترین باکتری های ایجاد کننده فساد غذایی است، به انسان وجود دارد. این بررسی به منظور تشخیص و بررسی الگوی مقاومت آنتی بیوتیکی باکتری اشریشیاکلی O157 جدا شده از گوشت قرقاول، کبک، اردک و غاز در استانهای گیلان، مازندران، اصفهان و فارس انجام شد.
مواد و روشها: برای انجام این بررسی به ترتیب تعداد 25، 17، 22 و 36 نمونه گوشت عضله سینه قرقاول، کبک، اردک و غاز از استانهای ذکر شده جمع آوری شد و در یخچال حاوی یخ سریعا به آزمایشگاه انتقال داده شد. نمونه ها ابتدا کشت داده شدند و سپس از کلنی های تیپیک که نشانگر اشریشیاکلی بودند، DNA استخراج گردید و واکنش زنجیره ای پلیمراز برای تایید تشخیص و شناسایی گروه سرمی O157 استفاده شد. در پایان الگوی مقاومت آنتی بیوتیکی به روش دیسک گذاری ساده مورد بررسی قرار گرفت.
یافته ها: نتایج نشان داد که 27% نمونه ها از نظر وجود اشریشیاکلی مثبت بودند که 6 نمونه آن دارای گروه سرمی O157 بودند. بررسیها نشان داد که بیشترین و کمترین فراوانی باکتری و گروه سرمی O157 به ترتیب متعلق به گوشت غاز و گوشت کبک بوده است. باکتری اشریشیاکلی جدا شده از گوشت طیور، بیشترین میزان مقاومت آنتی بیوتیکی را به سولفامتوکسازول و ونکومایسین و کمترین میزان مقاومت آنتی بیوتیکی را به جنتامایسین و سیپروفلوکساسین داشته است. اختلاف آماری معنی داری (05/0P<) بین میزان شیوع باکتری و گروه سرمی O157 بین گوشت غاز و کبک و همچنین بین میزان مقاومت به آنتی بیوتیک های مختلف دیده شد.
نتیجه گیری: پیشنهاد می شود که از روش دقیق، سریع و بی خطر واکنش زنجیره ای پلیمراز به منظور کنترل بهداشت گوشت طیور از نظر وجود پاتوژن هایی مثل اشریشیاکلی بهره گیری شود. این بررسی نشان می دهد که استفاده از روش دیسک گذاری ساده قبل از تجویز آنتی بیوتیک بسیار ضروری است.
Background and Aim: Despite the high value of poultry meat، there is no accurate control and inspection on poultry carcasses in the slaughterhouse. Therefore، the possibility of transmission of some bacteria like Escherichia coli which is one of the main causes of food poisoning is not unexpected. The present study was carried out to detect and analyse antibiotic resistance pattern of Escherichia coli O157 isolated from pheasant، partridge، duck and goose meat in Gilan، Mazandaran، Isfahan and Fars provinces، Iran.
Materials and Methods: For doing the experiments، 25، 17، 22 and 36 samples from pheasant، partridge، duck and goose chest muscle meat were collected، respectively. The samples were immediately transferred to the laboratory in cooler ice-pack. All samples were cultured and DNA was extracted from the typical bacterial colonies which represent Escherichia coli. Polymerase chain reaction was used to confirme the diagnosis and to detect O157 serogroup. Finally، the antibiotic resistance pattern was studied using simple disk diffusion method.
Results: The results showed that 27% of samples were positive for presence of Escherichia coli; 6 of them had the O157 serogroup. The goose and partridge meat had the highest and lowest frequencies of bacterium and O157 serogroup، respectively. The Escherichia coli which were isolated from poultry meat had the highest antibiotic resistance to sulfamethoxazol and vancomycinand the lowest antibiotic resistance to ciprofloxacinand gentamycin. A significant statistical difference (P < 0.05) was seen between the prevalence rate of the bacterium and O157 serogroup in goose and partridge meat and between the levels of resistance to various antibiotics.
Conclusion: We recommended using Polymerase Chain Reaction as an accurate، rapid and safe method to control presence of some pathogens like Escherichia coli. This study showed that using simple disk diffusion method is very essential before antibiotic prescription.
