چکیده:
اهداف: گاز خردل مادهای تاولزاست که برای اولینبار بهعنوان سلاح شیمیایی در جنگ جهانی اول و نیز در طول جنگ عراق علیه ایران مورد استفاده قرار گرفته است. هدف از این مطالعه بررسی ارتباط پلیمورفیسم C→T در جایگاه 509- پروموتر ژن TGF-β1 با عوارض دیررس ریوی در مصدومان شیمیایی با گاز خردل بود.
مواد و روشها: این مطالعه در میان 252 نفر جانباز شیمیایی که به دو گروه 107 نفر افراد مواجه با گاز خردل با مشکل ریوی و 145 نفر بدون مشکل ریوی تقسیم شده بودند، انجام شد. خون محیطی از افراد مورد مطالعه در لوله حاوی EDTA جمعآوری و سپس، DNA ژنومی از لوکوسیتها استخراج شد. برای تعیین ژنوتیپ افراد از روشPCR-RFLP استفاده شد. آنالیزهای آماری با استفاده از نرمافزار SPSS 18 و آزمون مجذور کای انجام گرفت.
یافتهها: در گروه مصدومان شیمیایی دارای مشکل ریوی 33 نفر و در گروه بدون مشکل ریوی 43 نفر دارای ژنوتیپ هموزیگوت CC بودند. 59 نفر از مصدومان شیمیایی دارای مشکل ریوی و 76 نفر از گروه بدون مشکل ریوی ژنوتیپ هتروزیگوت CT داشتند (967/ 0=p). همچنین، در گروه دارای مشکل ریوی 15 نفر و در گروه بدون مشکل ریوی 26 نفر دارای ژنوتیپ هموزیگوت TT بودند (473/ 0=p). تفاوت معنیداری بین توزیع ژنوتیپی مختلف پلیمورفیسم C→T در جایگاه 509- پروموتر ژن 1β-TGF در گروه مواجهه بدون مشکل و دارای مشکل ریوی مشاهده نشد.
نتیجهگیری: ارتباطی بین عوارض درازمدت ریوی ناشی از گاز خردل و پلیمورفیسم C→T در جایگاه 509- پروموتر ژن 1βTGF- وجود ندارد.
Aims: Sulfur mustard causes blister and was used for the first time as a chemical weapon in World War I and during the Iran-Iraq war as well. The aim of this study was to investigate the correlation of C→T polymorphism in -509 promoter region of TGF-β1 gene with long-term pulmonary complications in sulfur mustard chemical injures. Materials & Methods: This study was done on 252 chemically-injured veterans including 107 exposed subjects with pulmonary problems and 145 subjects without pulmonary problems. Peripheral blood samples were collected in the tubes containing EDTA. Then، genomic DNA was extracted of peripheral blood leukocytes. PCR-RFLP method was used to determine genotype of the subjects. Statistical analysis was done using SPSS 18 by Chi-square test. Findings: 33 subjects in chemically-injured patients with pulmonary problems group and 43 subjects in groups without pulmonary problems had CC homozygous genotype. 59 chemically-injured patients with pulmonary problems and 76 subjects of the group without pulmonary problems had CT heterozygous genotype (p=0.967). Also، 15 subjects of pulmonary problems group and 26 subjects of the group without pulmonary problems had TT homozygous genotype (p=0.473). No significant difference was observed between different genotype distribution of the polymorphism of C→T in -509 promoter region of TGF-β1 gene of with and without pulmonary problem groups. Conclusions: C→T polymorphism in -509 promoter region of TGF-β1 gene is not correlated with pulmonary problems in chemical injured veterans.
