چکیده:
سابقه و هدف: در حال حاضر پژوهش های بسیاری روی تولید نانوداروهای هدفمند جهت کاربرد آنها در درمان اختصاصی سرطان، انجام می گیرد. در این تحقیق، به منظور تولید نانودارویی با زیست سازگاری، اتصال و جذب سلولی بالاتر به عنوان عامل فوتوترمال تراپی هدفمند سرطان سینه، دو روش کونژوگاسیون مستقیم و غیرمستقیم، مقایسه گردید.
مواد و روش ها: نانومیله های طلا با طول موج جذبی 700 نانومتر طی دو روش مستقیم و غیر مستقیم به مولکول بومبسین، کونژوگه شدند. در روش مستقیم که روش متداول می باشد، پس از اتصال کوالان بومبسین به نانوذرات طلا، نانومیله ها پلی اتیلن گلیکوله گردیدند. در حالی که در روش غیر مستقیم، ابتدا نانومیله های طلا توسط پلی اتیلن گلیکول روکش شده و سپس مولکول های بومبسین توسط پیوند کوالان به نانوذرات متصل گردید. در مرحله ی آخر، دو کونژوگه ی حاصل از نظر پایداری در خون، زیست سازگاری، اتصال اختصاصی و جذب سلولی با یکدیگر مقایسه گردیدند تا بهترین روش، جهت تولید نانو دارویی با ویژگی های بهینه، به دست آید.
یافته ها: نتایج این تحقیق نشان داد هر دو نانو داروی تولید شده، پایداری بالا و مشابهی در خون دارند ولی نانو داروی تولیدی به روش غیرمستقیم، زیست سازگاری بالاتری از خود نشان داد. به علاوه، نانو داروی تولید شده به روش غیرمستقیم، حدود چهار برابر اتصال و جذب سلولی بالاتری نسبت به کونژوگه ی روش مستقیم از خود نشان داد.
نتیجه گیری: کونژوگاسیون غیرمستقیم، نانو دارویی با زیست سازگاری، اتصال و جذب سلولی بالاتر تولید کرده و روش مناسب تری جهت تولید نانو داروهای هدفمند می باشد.
Background and Aim: These nanomaterials are currently under investigation and developing for applications related to cancer therapy. To improve efficiency for targeted treatment of malignant tumors، we compare the two conjugation methods (direct and indirect) for production of bombesin-gold nanorods conjugate against gasterin release peptide receptor as a targeted photothermal therapy agent for breast tumor.
Materials and Methods: Bombesin conjugated with gold nanorods using the direct and indirect methods was used. The attachment of bombesin to gold nanorods and in vitro stability of two conjugates in mouse blood was studied by UV/vis spectroscopy. Cell toxicity for both of conjugate was investigated by MTT test. Selective binding and cellular uptake studies were performed with the T47D cell line. Results obtained of two conjugates compared to introduce best conjugation method.
Results: UV-vis spectroscopy confirmed the conjugation of bombesin with gold nanorods in two methods. The stability assessment proved same and high stability of two conjugates in mouse blood up to 4 h. Cytotoxicity study showed higher biocompatibility of indirect conjugate of bombesin and gold nanorods in comparison with its direct conjugate. In vitro studies showed that indirect conjugate of bombesin and gold nanorods have four-fold binding and cellular uptake in comparison with its direct conjugate.
Conclusion: These results show that the indirect conjugation was better than the direct conjugation to produce targeted nanodrug with low cytotoxicity، high binding and cellular uptake for photothermal therapy of breast cancer.
