چکیده:
گیاه تاتوره (.Datura metel L) گیاه دارویی مهمی است که جزو تیره سیب زمینی (Solanaceae) بوده و حاوی ترکیبات دارویی مهمی از جمله آتروپین و هیوسین (اسکوپولامین) می باشد که جزء تروپان آلکالوئیدها بوده و دارای خواص آنتیکولینرژیکی شناخته شده ای هستند. در سالهای اخیر القای بافت کالوس به عنوان روش جدیدی برای به دست آوردن متابولیتهای ثانویه مطرح شده است. هدف از این پژوهش، القای کالوس از برگ جدا شده گیاهچه های تاتوره میباشد تا بهترین غلظت هورمونی برای تولید کالوس بیشتر به دست آید. بذرهای تاتوره تحت شرایط استریل در محیط کشت MS کشت شدند و گیاهچه های تاتوره حاصل شدند. برگهای گیاهچه ها جدا شده و به محیط کشت MS بدون هورمون منتقل شد، اما کالوس زایی رخ نداد. در مرحله بعد برگهای گیاهچه ها در 9 تیمار هورمونی که حاوی غلظتهای مختلف اکسین 2,4-D و سیتوکینین کینیتین (Kin) بودند، به محیط کشت MS انتقال داده شدند. نمونهها چهار هفته در ژرمیناتور با دوره نوری 16 ساعت روشنایی و 8 ساعت تاریکی و دمای 25 درجه سانتیگراد قرار گرفتند. نتایج نشان داد حداکثر کالوسزایی در محیط کشت MS حاوی ترکیب دو هورمون با غلظتهای 1 میلیگرم بر لیتر 2,4-D و 1 میلیگرم بر لیتر Kin حاصل شد.
Datura metel L. is an important medicinal plant that is a part of the Solanaceae family and contains important pharmaceutical compounds, including atropine and hyoscine (scopolamine), which is part of the tropane alkaloids and has known anticholinergic properties. In recent years, callus tissue induction has been introduced as a new method for obtaining secondary metabolites. The aim of this study was to inoculate callus from the isolated leaves of Datura seedlings to obtain the best hormonal concentration for maximum callus production. Datura seeds were cultured in MS medium under sterile conditions and Datura seedlings were obtained. The leaves of the seedlings were isolated and transferred to the MS medium without hormone, but the callus did not occur. In the next step, the leaves of the seedlings were transferred to the MS medium in 9 hormonal treatments containing different concentrations of auxin 2,4-D and cytokinin kinetin (Kin). the samples with light period was 16 hours of light and 8 hours of darkness and temperatures of 25 ° C for four week. The results showed that maximum callus formation in MS medium containing two hormones with concentrations of 1 mg/L 2,4-D and 1 mg/L Kin was obtained.
