Abstract:
زمینه و هدف: ظهور کووید-19 توسط کروناویروس SARS-CoV-2 با علائم سندرم حاد تنفسی در ماه دسامبر سال 2019 در شهر ووهان کشور چین، با شیوع بسیار سریع همراه بوده و به سرعت به مرحله همهگیری جهانی رسید. این مطالعه با هدف معرفی انواع ژنتیکی ویروس مولد کووید-19 صورت گرفته است.
روش: بررسی فیلوژنتیکی ویروس SARS-CoV-2 به منظور شناسایی تنوع ژنتیکی ویروسهای شیوعیافته در ایران انجام و با 6 کشور منتخب مقایسه شد.
یافتهها: بررسی فیلوژنتیکی ویروس SARS-CoV-2 نشان داد که تنوعهای ژنتیکی ایجاد شده در ژنوم ویروس درحالحاضر به حدی نیست که بتواند اختلافات فیلوژنتیکی قابلتوجهی را بین ژنوم ویروسهای بهدستآمده از مناطق جغرافیایی گوناگون (در مجموع 19 کشور از قارههای مختلف) ایجاد کند ولی میتواند خوشهبندیهای متنوعی را شکل دهد. بررسی جهشهای ویروس SARS-CoV-2 نشان داد که ویروسهای شیوعیافته در ایران دارای شش تنوع ژنتیکی تکنوکلئوییدی (SNPs) در ژنهای غیرساختاری و ساختاری و یک اضافه شدن (Insertion) شش نوکلئوییدی در ناحیه ژن RNA-dependent RNA polymerase میباشند. مقایسه تنوعهای ژنتیکی ویروسهای ایران با شش کشور منتخب مورد بررسی (هند، آمریکا، نپال، ایتالیا، اسپانیا و فرانسه) نشان از منحصربهفرد بودن تنوعات ژنتیکی ویروسهای شیوعیافته در ایران دارد.
نتیجهگیری: نتایج مطالعات نشان میدهد که ارتباط مستقیمی بین جهشهای رخ داده در ژنوم ویروس SARS-CoV-2 و تغییر در بیماریزایی ویروس وجود دارد. بنابراین میتوان نتیجه گرفت که بررسی منظم تنوع ژنتیکی ویروس در هر کشور در طول زمان شیوع این بیماری همهگیر میتواند دادههای ارزشمندی را برای ساخت واکسن و دارو در اختیار قرار دهد.
Background:The incidence of COVID-19 by the coronavirus SARS-CoV-2 with symptoms of acute respiratory syndrome in December 2019 in Wuhan, China, has spread rapidly and change to a global epidemics.
Methods:SARS-CoV-2 virus phylogenetic study performed to identify the genetic diversity of the prevalent viruses in Iran and compared with 6 selected countries.
Results:Phylogenetic study of SARS-CoV-2 virus showed the genetic variations generated in the virus genome are not currently sufficient to distinguish significant phylogenetic differences between the genome of viruses obtained from different geographical areas (19 countries from different continents) but can form variety of clusters. A study of the SARS-CoV-2 virus mutations showed that the prevalent viruses in Iran had six single-nucleotide (SNP) genetic variations in non-structural and structural genes and one insertion of six nucleotides in the RNA-dependent RNA polymerase gene region. Comparing the genetic variations of viruses in Iran with the six selected countries (India, the United States, Nepal, Italy, Spain and France) indicate the uniqueness of genetic variations of the prevalent viruses in Iran.
Conclusion: Studies show there is a direct connection between mutations in the SARS-CoV-2 virus genome and changes in pathogenesis. Therefore, regular assessment of the virus genetic diversity in each country can provide valuable data for the development of vaccines and drugs.
