Abstract:
مقدمه: امروزه، استفاده از رنگهای طبیعی در مواد غذایی و دارویی اهمیت به سزایی دارد. فایکوسیانین به عنوان یک رنگدانه طبیعی با خاصیت آنتیاکسیدانی قوی از جلبک اسپیرولینا استخراج میگردد. به دلیل اثرات سمی گزارش شده از رنگهای سنتزی، دلایل مختلفی برای استفاده از رنگهای طبیعی در مصارف دارویی و غذایی وجود دارد.
مواد و روشها: در این تحقیق، تولید فایکوسیانین در شرایط مختلف دما (C˚ 30 و 35) ، شدت نور متغیر (0/2 و 5/3 کیلولوکس)، نوع منبع کربنی (گلوکز، اتانول و اسید استیک) و روش کشت (مداوم و غیر مداوم) برای تولید حداکثر فایکوسیانین مورد مطالعه قرار گرفت. آزمایشهایی مطابق با روش فاکتوریل کامل در دو شرایط ثابت و متغیر بر روی نمونهها انجام گردید.
یافتهها: نشان داد در روش غیر مداوم و نیمه مداوم غلظت منبع کربنی 0/1 میلی لیتر بر لیتر و نور 0/2 کیلولوکس با افزایش دما از30 به 35 درجه سلسیوس تولید فایکوسیانین در گلوکز نسبت به دو منبع کربنی دیگر با سرعت بیشتری کاهش پیدا کرد. با استفاده از غلظت 0/1 میلی لیتر بر لیتر منبع کربنی و نور 5/3 کیلولوکس، هر سه منبع در دمای30 درجه سلسیوس تفاوت معنی داری بین دو روش غیر مداوم و نیمه مداوم وجود نداشت.
نتیجهگیری: در هر دو روش به دلیل نور بالا مقدار تولید فایکوسیانین بالا بود. البته مقدار تولید با استفاده از گلوکز بیشترین مقدار بود و استفاده از اتانول نیز با اختلاف ناچیز میزان تولید خوبی را نشان داد.
Introduction: Nowadays the application of natural dyes in food and pharmaceutical products
are desirable and might be important. Phycociyanin is a natural pigment with powerful
antioxidant activity and is extracted from the Spirulina algae. Due to the possible toxic effects
of some synthetic colors and antioxidants the use of natural additives in food and
pharmaceutical product are perfected.
Materials and Methods: In this research, phycocyanin is produced at 30 and 35 , using
glucose, ethanol and acetic acid as carbon source in both batch and semi continues methods
with variable light intensitives of 2 and 3.5 Klx for maximum production. The experiments
were carried out according to Full Factorial method using fixed and variable conditions on the
samples.
Results: The results show that in batch and semi continues methods with 1.0 mlL-1 carbon
source and 2.0 Klx light intensity by increasing the temperature from 30 to 35 , the
production of plycociyanin in glucose as compared to the other two carbon sources have
rapidly declined. The results also indicated that by using a concentration of 1 mlL-1 carbon
source and light intensity of 3.5 Klx at 30 , there were not significant differences between
the batch and semi continuous methods.
Conclusion: It was concluded that the amount of production by using glucose was maximum
and also using ethanol showed good yields with minimal differences.
